Funny, this popped into my mind immediately, and I scrolled down and... bingo.
Now, I wouldn't call it the "greatest" but it may be one of the most:
recognizable?
catchy?
overplayed?
obvious?
I don't know. I'm at a loss for words.
Finally.

Eh? Black Magic Woman is a Peter Green riff - he wrote it, he played it, Santana merely covered it.

Enter Sandman is always what I immediately jump to when the term
greatest guitar riff ever
comes up. But since it's all personal opinion, it won't matter. I have probably changed my mind from time to time, and most people here hate that album, anyway. So it won't be a popular choice. Plus it's easy as hell to play, something many progheads scoff at.

that is why progheads are sick and twisted...
it wouldn't be a great guitar riff if it wasn't easy to play.. or more to the point.. if it was so hard to play. Why do they become so great... first they 'grab you' and have a life their own..much of that stems from power.. not frickin complexity and because of that.. everyone of the true greatest riffs is that way because every teenager who has ever grabbed a guitar has learned the riff. They don't become great because some geek who thinks it has to be complex to be great... they become great when they resonate with everyone.. and that means the 98% of music lovers that are not hung up on difficulty or complexity

I thought about Iron Maiden too with 'Ghost of The Navigator' 'Rime of the Ancient Mariner' and the incredible 'Brave New World' all classic riffs. As far as easy to play riffs. Sabbath's 'Paranoid', 'The Wizard' and 'Snowblind' are phenomenal.
Have to mention Metallica's 'Master of Puppets', 'For whom the bell tolls' and 'Sad But True'. Awesome power riffs.
And I am glad someone mentioned 'Aqualung' as that is an amazing riff.

Some bands would kill just to have one of Dave Brock's riffs: The Riff machine was so rifftastic and riffmongous that he would get more riffs in one 'song' than others had in a lifetime.
